Laboratory research focuses on how genes and molecules orchestrate the development of an embryo. The knowledge obtained may help to discover new molecules and specific gene/cell treatments to prevent and cure diseases affecting mankind both in the adult and embryonic stages, as well as inducing endogenous in vivo regenerative responses that may allow for tissue and organ regeneration. The McVicker laboratory studies how human genetic variation affects gene regulation by combining experimental approaches with computational analyses. We are especially interested in identifying pathogenic regulatory variants that act in immune cells and cancer cells. Currently, we are developing new tools to discover somatic regulatory mutations in cancer genomes, and manipulating cell lines with high-throughput CRISPR technologies to discover new regulatory sequences. In much of our research, we develop sophisticated computational and statistical methods so that we can extract subtle signals from noisy experimental data. The research focuses on important problems related to cancer initiation, detection, progression, metastasis and treatment. These projects are based on our recent observations concerning cell state plasticity that arises during cancer progression. Specifically, we are using a single cell RNA sequencing, epigenetic profiling, and developing novel in vivo and in vitro strategies involving mouse models and organoid cultures to determine how cancer cells undergo developmental reprogramming to generate intra-tumoral heterogeneity and metastasis. We use computational methods based on mouse cancer models and human databases. The Chalasani lab at Salk is seeking ambitious and motivated post-doctoral fellows to join their Sonogenetics team. The goal of this project is to develop a new non-invasive method to manipulate neurons and non-neuronal cells ( www.sonogenetics.salk.edu ).
